Improvement of single-cell gel electrophoresis (SCGE) alkaline comet assay

چکیده
Although the single-cell gel electrophoresis (SCGE) alkaline comet assay can detect DNA damage quickly and with high sensitivity, it does not work well for cryopreserved sperm of some marine teleosts. Using large yellow croaker Pseudosciaena crocea sperm that were cryopreserved in increasing concentrations (5 to 30%) of DMSO, we made modifications to the classic SCGE method that included using common microscopic slides without rough surfaces, pretreatment of sperm before gel spread, and a single layer of gel. Electrophoresis conditions were 130 mA, 15 V, and 60 min, providing a high SCGE sensitivity and definition of the comet image. The improved method is useful for detecting cryopreserved sperm DNA damage of some marine teleosts, especially for testing the quality of genetic resources.

Comparison of the In Vitro Mutagenicity of Artemisia draconculus L. With Sodium Dichromate by Performing Single Cell Gel Electrophoresis (SCGE) or the Comet Assay
BACKGROUND The increasing use of herbal drugs and their easy availability have necessitated the use of mutagenicity tests to analyze their toxicity and safety. OBJECTIVES The aim of this study was to determine the in vitro mutagenicity of Artemisia draconculus L., a herbal drug, by performing single cell gel electrophoresis (SCGE). MATERIALS AND METHODS In this study, we obtained a herbal d...
متن کاملUse of single cell gel electrophoresis (comet assay) modifications for analysis of DNA damage.
Single cell gel electrophoresis (SCGE) or comet assay is a rapid and sensitive fluorescent microscopic method which allows measurement of DNA strand breaks in individual cells. Modifications of SCGE conditions permitted to detect different types of DNA damage. In order to characterize DNA damage induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and hydrogen peroxide (H2O2) in Chinese hamst...
متن کاملSingle-cell gel electrophoresis assays with human-derived hepatoma (Hep G2) cells.
The purpose of the present study was the development of a protocol for detecting chemically-induced DNA damage, using the alkaline single-cell gel electrophoresis (SCGE) assay with human-derived, metabolically competent hepatoma (Hep G2) cells. Previous studies indicated that Hep G2 cells have retained the activities of certain phase I and phase II enzymes and reflect the metabolism of genotoxi...
متن کاملDNA Damage Induced by Tartrazine in rat whole blood using Comet Assay (Single Cell Gel Electrophoresis)
Tartrazine is an orange-colored, water soluble powder widely used in food products, drugs and cosmetics. The acceptable daily intake (ADI) oh human is 0-7.5 mg/kg body weight. The objective of our study was to examine the genotoxic effect of different Tartrazine concentrations of fresh whole blood cells; by using alkaline single-cell gel electrophoresis (SCGE), also called the comet assay. This...
متن کاملEvaluation of in vivo genotoxicity of cypermethrin in Drosophila melanogaster using the alkaline Comet assay.
The single cell gel electrophoresis (SCGE) assay, also known as the Comet assay, is one of the most promising genotoxicity tests developed in recent years to measure and analyse DNA damage in single cells. The present study was undertaken to assess the in vivo genotoxicity of the synthetic pyrethroid cypermethrin in brain ganglia and anterior mid gut of Drosophila melanogaster. Freshly emerged ...
